Usage
Amlodipine inhibits the influx of calcium ions into vascular smooth muscle and cardiac muscle. Through this mechanism, amlodipine causes vasodilation of the vascular smooth muscle which results in a reduction in blood pressure. Amlodipine has not been found to affect serum calcium concentrations.

Dosage
Norvasc is the besylate salt of the long-acting calcium channel blocker amlodipine. Amlodipine besylate is found as a white crystalline powder. Norvasc tablets are white tablets that are available in doses of 2.5 mg, 5 mg and 10 mg. Depending on the dosing, amlodipine tablets are diamond, round, or elongated octagon in shape. Norvasc tablets should be stored at controlled room temperatures of 59° to 86° F.

Amlodipine – Metabolism
After oral administration, peak plasma concentrations of Norvasc (amlodipine) occur between 6 and 12 hours. The bioavailability is not affected if taken with food. While circulating, more than 90% of amlodipine is bound to proteins in the plasma. Amlodipine is converted into inactive metabolites in the liver and the majority of amlodipine metabolites are excreted in the urine. Amlodipine has a terminal elimination half-life of 30-50 hours which is not significantly affected in the setting of renal impairment.
