Malarone consists of a combination of two antimalarial agents – atovaquone and proguanil hydrochloride. Atovaquone is insoluble in water and is found as a yellow crystalline solid. Proguanil HCl is slightly soluble in water and is a white crystalline solid.
Malarone – Side Effects
Malarone, like other medications, can cause side effects. Side effects from Malarone use include, but are not limited to, abdominal pain, nausea, vomiting, headache, diarrhea and anorexia. Malarone can interact when used with other drugs and should be used with caution in those instances. Malarone should not be used in people with renal impairment. Malarone is contraindicated in people with a known hypersensitivity or prior allergic reaction to atovaquone, proguanil hydrochloride or any component of the formulation. Malarone and alcohol should not be used together. Concerns about malarone’s side effects should be directed to one’s medical provider before use of the medication.

The component drugs in Malarone work by interfering with processes necessary for the synthesis of DNA in the parasite. Both atovaquone and proguanil hydrochloride are mostly bound to proteins. The elimination half life of atovaquone is 2 to 3 days and that of proguanil hydrochloride is 12 to 21 hours in adults. Because atovaquone has some susceptibility to dietary fat, Malarone should be taken with food or a milky drink. Malarone tablets are oral medications available in different forms – atovaquone 250 mg and 100 mg proguanil hydrochloride tablets and atovaquone 62.5 mg and 25 mg proguanil hydrochloride tablets. Malarone tablets are pink and should be stored at 25° C.
