What is ondansetron? Ondansetron, marketed under the brand name Zofran, is a medication indicated for the prevention of nausea and vomiting. Specifically, ondansetron is indicated for postoperative nausea and vomiting, nausea and vomiting associated with radiotherapy in certain classes of patients, and nausea and vomiting associated with moderate or highly emetogenic cancer chemotherapy. How ondansetron works is not fully understood. Ondansetron (Zofran) is a selective 5-HT3 receptor antagonist and, because serotonin receptors are present all over the body, it is possible that ondansetron works both centrally and peripherally.
Ondansetron – Side Effects
Ondansetron and Zofran should not be used by anyone with a known hypersensitivity or allergy to the medication or any of the components or by anyone who is using apomorphine. Common adverse events in adults using ondansetron include headache, fatigue, diarrhea and constipation. Zofran (ondansetron) also has other significant side effects, including, but not limited to, QT interval prolongation, serotonin syndrome and masking of progressive ileus following surgery or chemotherapy. No antidote has been identified for ondansetron overdoses. Ondansetron as Zofran is available in the form of tablets, orally disintegrating tablets (ODT) and oral solution. When found in tablet or ODT form, Zofran is available in 4 mg and 8 mg dosages. As an oral solution, Zofran is available in a 4 mg / 5 mL suspension. Zofran tablets are oval, film-coated tablets that are white or yellow in color. Zofran ODTs are white, round, plano-convex tablets. Zofran oral solution is a clear, colorless to light yellow liquid with a strawberry odor. Zofran tablets contain the active ingredient ondansetron hydrochloride as the dihydrate. As a racemic form of ondansetron, ondansetron hydrochloride selectively blocks serotonin 5-HT3 receptor types. Zofran ODTs contain ondansetron base, another racemic form of ondansetron that works in a similar way to ondansetron hydrochloride. Approximately 56% of ondansetron is available following oral administration of an 8 mg tablet and ondansetron metabolites are found in the urine.
